Done this week

  • Added a beam expander to the PMT path to make up for the beam shrinkage due to the relay lenses (-30mm and 200mm lens)
  • Added mirrors, lenses and dichroics
  • Positioned relay/beam expander lenses and adjusted the beam path to be the appropriate length based on these positions.
  • Cleaned up the the microscope design by stabilizing various parts, decreasing distances where possible, straightening cage rods, etc.
  • The galvos were not working, so we trouble shot the code and found that we were missing a "start" command at the end of the code. All fixed!
  • In order to align the laser path to the galvos, we adjusted the height of the beam path and added a second mirror for additional flexibility.

To do next week

  • Get a bright field image and align the laser so we can see it
  • Position lenses in PMT beam path
  • Install the pinhole and PMT and get a confocal image of a fluorescent slide!
  • Install new laser and beamsplitter/photodiode system

Risks and Issues

  • We're a little bit behind schedule!
  • The laser-->galvos beam path isn't 100% straight
  • What is the best way to normalize an image with the photodiode data?
