Making PSF slides

  • Use PS-Speck ORANGE beads, 170nm
  • Using a small pipette tip, spread 2-3uL of beads around to the size of a 18x18 cover slip (do not draw a circle with permanent marker, as its drying seems to mess up the bead distribution)
  • Let the slides air dry
  • Drop ~5uL glycerol on the slide where the beads are
  • Cover gently with cover slip, allow glycerol to spread to edge of cover slips
  • Seal edges with nail polish
